Can I assume SATA Hard Drive Bay Adapter III (43N3412) support SATA III ? I could not find the detail specification for this adapter.

The adapter doesn't matter, but t420s does not have an optical Sata interface with III criteria, so you'd better use the main hard drive for faster speed.

Also, I don't think Lenovo makes any SATA 3 bay adapters.
