I'm considering buying an X220 and have a question:
If I use an SSD as my system drive, and install a HDD for storing movies and large data files, will that HDD spin when I'm not reading/writing to it? In other words, if I'm not accessing the HDD will it still draw enough power that it would noticeably shorten battery life when compared to an SSD-only setup?

Unfortunate yes , hdd draws some power to keep controler ready-to-start mode
In my oppinion the best solution to save power would be install msata ssd for OS and apps and SSD for data
