I didn't pay attention until today (and can't report any side effects), but here goes;
I downloaded the latest Access Connections installer.
The default directory that the installer unpacks the data is C:\Drivers\W2K\Accon
I checked that folder and see that there are still the files of the previous version (4.21).
Is it safe to delete them (manually) before I let the install package unzips the files there?

It is safe to delete them, but it might be best to uninstall Access Connections first. FYI, your profiles are not stored in that directory, so do not worry about losing them by deleting the files in that directory.
This is what I do. I uninstall whatever I am upgrading, delete the files in the installation folder, then extract the upgraded files to the installation folder (by running the executable I downloaded). I have seen sometimes, if you do not delete the files, Software Installer will report that that software is still on the machine, although perhaps uninstalled.
